Cas9 Cell Lines

Cas9 stable cell line (i.e. Cas9 expressing cells), is the cell line that transduced with the lentivirus carrying the Cas9 nuclease gene. After antibiotic resistance screening, a cell line that can stably express Cas9 nuclease is generated. Cas9 cell line can further simplify the CRISPR/Cas9 mediated gene-editing experiments.

Why use Cas9 stable cell line?

-sgRNA library screening: Generally, CRISPR libraries have 1 vector system and 2 vector system. Because of the large size of the Cas9 sequence and the loading limitation of lentivirus vectors, it is very difficult to ensure the high titer of the 1 vector system library when packaging the virus. In this case, the 2 vector system has the advantage. Using the pre-made Cas9 stable cell line, the trouble of simultaneous transduction of Cas9 lentivirus and gRNA lentivirus can be avoided. The lentivirus packaged with gRNA can directly infect Cas9 stable cell line, to achieve library screening. This method can reduce the difficulty of the experiment, and increase screening efficiency.

-High editing efficiency: Simply transfect sgRNA into Cas9 cell line to achieve gene knockout. And gene knock-in can be achieved by transfection of sgRNA and donor DNA. Using Cas9 stable cell line can bypass the problem of low transfection efficiency of RNP ribonucleoprotein complex macromolecules, and significantly improve gene editing efficiency.
