▍  CRISPR Library Classification

The CRISPR whole genome sgRNA library can knockout any human or mouse genes and non-coding genes, such as miRNA genes. Each sgRNA is cloned into the optimized lentiviral vector to achieve efficient transduction of primary cells or cell lines. The cells should be transduced by pooled lentiviral library with low MOI to ensure that no more than 1 sgRNA gets in each cell. After transduction and before screening, NGS should be performed to evaluate the sgRNAs performance in cells. After screening, another round of NGS and data analysis is needed to identify the sgRNA that is missing or enriched during the screening process.

Sub-pool library targets at genes involved in some specific pathways or biological processes. Customized sgRNA library for CRISPR screening is an important means in functional genomics research.
